PSYC 373 Lab Report 1: Action Potential Simulation 18 points

This lab report will allow you to apply your learning about the resting potential, action potentials, and postsynaptic potentials to a simulation of neuronal conduction. Complete this lab report after you have watched the Neurophysiology videos. Refer back to the videos/slides as needed to answer the questions. You may also find the Metaneuron manual (available on the website) to be helpful. Experiment 1: Messing with membrane parameters at resting potential If you have not already done so, go to www.metaneuron.org to open the program. Download the MetaNeuron program and go through the prompts. Make sure you are downloading the correct version for your operating system. Once MetaNeuron has opened, go to the top of the window and select “Lesson”. Select “Lesson 1: Resting Membrane Potential”. This lesson demonstrates how K+ and Na+ channels contribute to the generation of resting membrane potential. 1. Record the concentrations (in mM) of sodium and potassium on the inside and outside of the cell, respectively. Which is found at a greater concentration inside the cell? Which is found at a greater concentration outside the cell? (Hint: consider the membrane permeability for sodium vs. potassium.) Record tshe value of the membrane potential in mV. (3 pts.) 2. The equilibrium potential of a neuron is the point at which there is no net flow of ions across the cell membrane. Set the concentrations of Na+ out, Na+ in, K+ out, and K+ in to 100. What happens to the value of the equilibrium potential for Na+ and K+? What happens to the value of the membrane potential? Explain why these changes happen. (4 pts.) Experiment 2: Messing with action potentials Go to “Lesson” and select “Lesson 4: Axon Action Potential”. This lesson demonstrates how voltage-dependent and time-dependent Na+ and K+ conductances generate action potentials. The Na+ conductance represents Na+ channels, and the K+ conductance represents the delayed K+ channels. 1. Change the Amplitude of Stimulus 1 to 30 uA. What is the value of the membrane potential? (Look at the yellow line on the graph to determine the approximate value). What is the threshold stimulus amplitude (in mA) for initiating an action potential? (The threshold stimulus is best measured by adjusting the stimulus amplitude in 5 mA increments until it is just large enough to initiate an action potential. The lowest value at which an action potential can be generated is the threshold stimulus amplitude.) (2 pts.) 2. The symptoms of multiple sclerosis are caused by action potential conduction failure due to the demyelination of CNS axons. The symptoms of multiple sclerosis are alleviated by lowering a patient’s core body temperature while symptoms are exacerbated by an exercise-induced rise in body temperature. Change the Amplitude of Stimulus 1 to 60 uA. Lower the Temperature by 1 degree at a time. How much does the temperature have to be lowered to evoke an action potential? Keep lowering the temperature. At what temperature does the membrane potential return to the resting value? What does this tell us? (3 pts.) Experiment 3: Postsynaptic Potentials Go to “Lesson” and select “Synaptic Potential and Current” 1. When you first open the lesson, the graphs are set to depict a postsynaptic neuron onto which an excitatory neurotransmitter (e.g., glutamate) was released. The graph all the way to the left (yellow line) represents the value of the membrane potential. When the initial (default) settings are in place, does this graph depict an EPSP or IPSP? Explain your answer. Record the current values for Na+ permeability, K+ permeability, and Cl- permeability. Which is highest? (4 pts.) 2. Change the synapse type to “fast inhibitory synapse”. This depicts a synapse where an inhibitory neurotransmitter (e.g. GABA) has been released. Change the value for Clpermeability to 10 and the value of the Holding Current to 20 uA. What happens to the graph all the way to the left (yellow line)? Does this graph show an EPSP or IPSP? (2 pts.)
